이승재 건대교수 퇴행성뇌질환 확산원리 규명
뇌질환 치료ㆍ진단법 개발 새 길 열어


(서울=연합뉴스) 김영섭 기자 = 파킨슨병과 치매도 광우병과 마찬가지 방식으로 단백질 변성체의 신경세포 간 이동에 의해 뇌 손상부위가 늘어난다는 연구결과가 나왔다.


건국대 의생명과학과 이승재 교수는 건국대 의대 이혜진 교수, 미국 샌디에이고 캘리포니아대학(UCSD) 엘리에저 마슬리아 교수와의 공동연구를 통해 일부 뇌 영역에서 시작된 뇌질환이 변성된 단백질의 신경세포 간 전파에 의해 여러 뇌 부위로 확대된다는 사실을 세계 최초로 규명했다고 28일 밝혔다.


이번 연구는 파킨슨병이나 치매 등 뇌질환의 체내 진행원리가 광우병 등의 프리온(prion)병과 유사성이 있음을 밝힌 획기적 성과로서 퇴행성 질환의 진행 원리에 관한 새로운 패러다임을 제공했다는 평가다.


이 교수팀의 연구결과는 세계적 과학 권위지인 미국립학술원회보(PNAS) 인터넷판 28일자에 발표됐다.


이 교수팀은 뇌질환 모델 세포와 그렇지 않은 세포를 함께 배양하는 공배양 세포연구 방법과 정상적인 생쥐에서 신경줄기세포를 얻어 뇌질환에 걸린 생쥐의 뇌에 이식하는 방법 두 가지를 사용했다.


이는 세포배양 방식과 생체 내에서의 반응을 모두 알아보기 위해서라고 연구진은 설명했다.
이 결과 ‘알파-시뉴클린(alpha-synuclein)’이란 신경세포의 단백질이 변성된 후 신경세포로부터 분비돼 인접 신경세포로 전이되고 또한 전이된 단백질에 의해 신경세포의 사멸이 유도된다는 사실이 두 연구방식 모두에서 밝혀졌다.


연구진에 따르면 파키슨병 등 퇴행성 뇌질환의 발병 초기에 손상된 신경세포(뉴런)에서 정상 알파-시뉴클린 단백질(α-syn monomer)이 변성을 일으켜 변성체(α-syn aggregate)를 형성한다.


이어 이들 변성체는 신경세포 밖으로 분비되고 나서 인접 신경세포로 이동해 정상 단백질의 변성을 유도하고, 단백질 변성이 인접 신경세포에서 증폭되는 현상이 반복됨으로써 뇌의 여러 부위로 질병이 확산되는 것으로 나타났다.


뇌 세포 사이의 신경전달을 돕는 단백질로 퇴행성 뇌질환의 주요 원인 물질인 알파-시뉴클린의 변성은 파킨슨병과 치매의 발병 기전(메커니즘)에 중요한 역할을 한다고 알려져 있다.


이번 연구는 일부 신경세포에서 발생한 단백질의 변성이 뇌의 여러 부위로 퍼져 병리현상의 확산이 일어날 수 있음을 보여줬다는 데 의의가 있다.


특히 단백질 변성체에 의한 질병의 진행 및 확산은 광우병 등의 프리온병에 국한된 현상이라고 여겨져 왔으나, 이번 연구를 통해 프리온의 확산 원리가 파킨슨병과 치매 등의 여러 퇴행성 뇌질환에 광범위하게 적용될 수 있는 공통 원리라는 실험적 근거를 제시했다.


퇴행성 뇌질환과 관련된 단백질 변성체는 정상 단백질을 변성시킴으로써 복제가 가능한데, 이는 광우병을 비롯한 프리온병의 감염 원리를 설명하는 기전으로 알려져 있다.


알츠하이머병과 파킨슨병을 포함한 퇴행성 뇌질환은 60, 70세 이상의 노년층에서 주로 발병하는 대표적 노인성 질환으로 날로 고령화하는 우리 사회에 커다란 부담으로 대두되고 있다.


이 질병들의 발병 원인은 아직 명확히 밝혀지지 않았으며 여러 가지 이론이 제시돼 연구 중이다.


최근 가장 중요하게 부각되는 이론은 신경세포에 있는 특정 단백질의 구조적 변성 및 응집에 의해 신경세포가 손상되고 결국 사멸된다는 것이다.


파킨슨병 및 치매 등의 노인성 뇌질환은 병이 진행될수록 점점 많은 뇌 부위의 손상을 일으키고 점차로 확대되면서 다양한 증상을 보인다.


이 교수는 “이번 연구는 변성 단백질을 손쉽게 검출하고 변성 단백질이 다른 세포로 이동하는 과정을 차단하는 방법 등을 통해 향후 뇌질환 진단 및 치료법 개발에 응용될 수 있을 것으로 기대된다”고 밝혔다.


한편, 미학술원회보는 이 교수팀 연구의 중요성을 인식해 프리온의 전파 원리를 규명해 1997년 노벨 생리의학상을 수상한 스탠리 프루시너 교수의 해설과 주석을 함께 실어 학계에 상세히 보고할 계획이다.

Abstract

Neuronal accumulation of α-synuclein and Lewy body formation are characteristic to many neurodegenerative diseases, including Parkinson’s disease (PD). This Lewy pathology appears to spread throughout the brain as the disease progresses. Furthermore, recent studies showed the occurrence of Lewy pathology in neurons grafted into the brains of PD patients, suggesting the spread of pathology from the host tissues to the grafts. The mechanism underlying this propagation is unknown. Here, we show that α-synuclein is transmitted via endocytosis to neighboring neurons and neuronal precursor cells, forming Lewy-like inclusions. Moreover, α-synuclein was transmitted from the affected neurons to engrafted neuronal precursor cells in a transgenic model of PD-like pathology. Failure of the protein quality control systems, especially lysosomes, promoted the accumulation of transmitted α-synuclein and inclusion formation. Cells exposed to neuron-derived α-synuclein showed signs of apoptosis, such as nuclear fragmentation and caspase 3 activation, both in vitro and in vivo. These findings demonstrate the cell-to-cell transmission of α-synuclein aggregates and provide critical insights into the mechanism of pathological progression in PD and other proteinopathies.
